The anti-MAPK1 pH-dependent antibody (clone VB22B) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Mitogen-activated protein kinase scaffold protein 1 (MAPK1). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MAPK1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MAPK1 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MAPK1, also known as ERK2, is a key kinase in the MAPK/ERK signaling pathway that regulates cell proliferation, differentiation, and survival.
