The anti-NTRK3 pH-dependent antibody (VS-0825-YC275) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human NTRK3. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for NTRK3 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from NTRK3 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human NTRK3, or TrkC, is a receptor tyrosine kinase that binds to Neurotrophin-3 (NT-3). It is essential for neuronal survival and differentiation, and oncogenic fusions of NTRK3 are a driver of a variety of cancers.
