The anti-SLAMF7 pH-dependent antibody (VS-0825-YC308) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human SLAMF7. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SLAMF7 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SLAMF7 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human SLAMF7 (SLAM Family Member 7), also known as CD319, is a transmembrane protein expressed on natural killer (NK) cells and plasma cells. It is involved in NK cell activation and is a key therapeutic target for the treatment of multiple myeloma.
