- Introduction
- T cell immunoreceptor with Ig and ITIM domains (TIGIT) is an immune checkpoint receptor expressed on T cells, NK cells, and Tregs. It binds ligands CD155 (PVR) and CD112 (Nectin-2) on tumor cells or APCs, inhibiting T cell and NK cell cytotoxicity and promoting immune evasion. Overexpressed in various cancers (melanoma, NSCLC, RCC) and associated with poor prognosis, TIGIT is a promising next-generation checkpoint target. Inhibitors (tiragolumab, vibostolimab) are being tested alone or in combination with PD-1/PD-L1 blockers, showing synergistic anti-tumor activity by simultaneously targeting multiple immune checkpoints.
