BAD: A Sentinel of the Mitochondrial Apoptosis Pathway

BCL2 Associated Death Promoter (BAD) is a pro-apoptotic BH3-only member of the BCL2 family. BAD promotes apoptosis in its dephosphorylated state by binding and sequestering anti-apoptotic proteins, such as BCL2 and BCL-xL, and thereby disinhibits the function of the pro-apoptotic BAX and BAK proteins which permeabilize the outer mitochondrial membrane. BAD is under strict regulatory control by phosphorylation. Cell survival signals, mediated by PI3K and subsequent activation of AKT and other kinases, phosphorylate BAD at specific serine residues. Phosphorylated BAD, in turn, provides a binding site for 14-3-3 proteins, which sequester BAD to the cytoplasm and prevent it from exerting its pro-apoptotic function.

Background

The protein encoded by this gene is a member of the BCL-2 family. The BCL-2 family members are regulators of programmed cell death. It positively regulates cell apoptosis by forming heterodimers with BCL-xL and BCL-2, and reversing their death repressor activity. Phosphorylation regulates proapoptotic activity of this protein. Protein kinases AKT and MAP kinase, as well as protein phosphatase calcineurin were found to be involved in the regulation of this protein. Alternative splicing of this gene results in two transcript variants which encode the same isoform.

The Ser-75/Ser-99 phosphorylated form binds 14-3-3 proteins (By similarity). Interacts with AKT1 and PIM3. Interacts (via BH3 domain) with NOL3 (via CARD domain); preventing the association of BAD with BCL2 (By similarity). Interacts with HIF3A (via C-terminus domain); the interaction reduces the binding between BAD and BAX (By similarity). Interacts with GIMAP3/IAN4 and GIMAP5/IAN5 (PubMed:16509771).
