CAMK2B: A Kinase with Structural Roles at the Synapse

Calcium/Calmodulin-Dependent Protein Kinase II Beta (CAMK2B) is one of the two major isoforms of CAMKII in the brain, and it co-assembles with the alpha isoform into hetero-dodecamers. As with CAMK2A, this protein is a calcium-activated serine/threonine kinase that plays critical roles in mediating synaptic plasticity, with major effects on the structural plasticity of dendritic spines. CAMK2B has a unique linker region that enables binding directly to F-actin, providing a direct structural role in anchoring the kinase complex at the synapse and the regulation of dendritic spine morphology. Mutations in this gene have been identified as the cause of neurodevelopmental disorders, leading to intellectual disability and seizures.

Background

The product of this gene belongs to the serine/threonine protein kinase family and to the Ca(2+)/calmodulin-dependent protein kinase subfamily. Calcium signaling is important for several aspects of plasticity at glutamatergic synapses. The enzyme is likely to be a tetramer in mammalian cells and may be composed of four different chains (alpha, beta, gamma and delta). The product of this gene is one of the beta chains. There is a possibility that distinct isoforms of this gene have different cellular localizations and may interact differently with calmodulin. Multiple transcript variants have been found due to alternative splicing.
