Creative Biolabs, your professional partner in the field of immunology and cancer biology, is offering you highly specific anti-CASP8 recombinant antibodies. Caspase-8 (CASP8) is an initiator caspase of the extrinsic pathway of apoptosis, which is considered a primary link between cell surface death receptors and the execution machinery of cell suicide. High-quality antibodies for this enzyme are crucial for studying TNF family signaling, immune surveillance, and chemoresistance mechanisms.

CASP8: The Initiator of Death Receptor Signaling

Caspase-8 (CASP8) is an initiator cysteine protease, which is involved in the extrinsic pathway of apoptosis. It is recruited to the Death-Inducing Signaling Complex (DISC) through its death effector domains (DEDs). DISC is assembled on the cytoplasmic side of cell surface death receptors (Fas or TRAIL receptors) upon ligand binding. The recruitment of multiple pro-caspase-8 molecules to DISC brings them in close proximity, resulting in their dimerization and auto-activation. Active caspase-8 subsequently initiates the apoptotic signaling cascade by direct cleavage and activation of downstream executioner caspases (e.g., caspase-3). Caspase-8 can also cleave the protein Bid and link to the intrinsic mitochondrial pathway of apoptosis.

Alternative Names

CASP8; caspase 8, apoptosis-related cysteine peptidase; CAP4; MACH; MCH5; FLICE; ALPS2B; Casp-8; caspase-8; FADD-like ICE; MACH-alpha-1/2/3 protein; apoptotic protease Mch-5; MACH-beta-1/2/3/4 protein; apoptotic cysteine protease; ICE-like apoptotic protease 5; MORT1-associated ced-3 homolog; FADD-homologous ICE/CED-3-like protease; caspase 8, apoptosis-related cysteine protease

Background

This gene encodes a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes composed of a prodomain, a large protease subunit, and a small protease subunit. Activation of caspases requires proteolytic processing at conserved internal aspartic residues to generate a heterodimeric enzyme consisting of the large and small subunits. This protein is involved in the programmed cell death induced by Fas and various apoptotic stimuli. The N-terminal FADD-like death effector domain of this protein suggests that it may interact with Fas-interacting protein FADD. This protein was detected in the insoluble fraction of the affected brain region from Huntington disease patients but not in those from normal controls, which implicated the role in neurodegenerative diseases. Many alternatively spliced transcript variants encoding different isoforms have been described, although not all variants have had their full-length sequences determined. Interaction

Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 18 kDa (p18) and a 10 kDa (p10) subunit (PubMed:10508784). Interacts with FADD, CFLAR and PEA15 (PubMed:10442631). Interacts with TNFAIP8L2 (By similarity). Interacts with CASP8AP2 (PubMed:16378960). Interacts with RFFL and RNF34; negatively regulate CASP8 through proteasomal degradation (PubMed:15069192). Interacts with NOL3; decreases CASP8 activity in a mitochondria localization- and phosphorylation-dependent manner and this interaction is dissociated by calcium (PubMed:15509781).
