PRKAA1: A Key Therapeutic Target for Multiple Diseases due to Its Role in Metabolic Regulation
PRKAA1, a serine/threonine kinase acting as a central hub in cellular signaling, plays a critical role in multiple pathological conditions by governing key biological processes linked to disease progression. In metabolic syndromes, this enzyme functions as a guardian of energy balance, orchestrating pathways that regulate glucose uptake, fatty acid oxidation, and mitochondrial function. When its activity is disrupted, cells fail to maintain optimal energy homeostasis, leading to insulin resistance, dyslipidemia, and the subsequent development of type 2 diabetes and obesity. In oncology, it influences tumor biology through dual mechanisms: under energy-deprived conditions, it promotes autophagy to sustain cancer cell survival, while in nutrient-rich environments, it can enhance anabolic pathways that drive uncontrolled proliferation. Such context-dependent roles make it a complex target in cancer therapy, as its modulation may either inhibit tumor growth or inadvertently support malignant progression. In neurodegenerative disorders, its dysfunction impairs axonal energy transport and disrupts the clearance of misfolded proteins, contributing to synaptic degeneration and the accumulation of toxic aggregates characteristic of Alzheimer’s and Parkinson’s diseases. Additionally, in cardiovascular diseases, altered activity of this kinase has been linked to myocardial energy deficit, compromising cardiac contractility and accelerating heart failure. By integrating signals from nutrient availability, stress responses, and cellular metabolism, this molecular regulator bridges multiple disease domains, highlighting its potential as a therapeutic target for interventions that address the underlying metabolic and signaling aberrations across diverse pathologies.
Alternative Names
Complement PRKAA1; AMPK1; 5'-AMP-activated protein kinase catalytic subunit alpha-1; AMPK subunit alpha-1; Acetyl-CoA carboxylase kinase; ACACA kinase; Hydroxymethylglutaryl-CoA reductase kinase; HMGCR kinase; Tau-protein kinase PRKAA1
Background
The protein encoded by this gene belongs to the ser/thr protein kinase family. It is the catalytic subunit of the 5'-prime-AMP-activated protein kinase (AMPK). AMPK is a cellular energy sensor conserved in all eukaryotic cells. The kinase activity of AMPK is activated by the stimuli that increase the cellular AMP/ATP ratio. AMPK regulates the activities of a number of key metabolic enzymes through phosphorylation. It protects cells from stresses that cause ATP depletion by switching off ATP-consuming biosynthetic pathways. Alternatively spliced transcript variants encoding distinct isoforms have been observed.

AMPK is a heterotrimer of an alpha catalytic subunit (PRKAA1 or PRKAA2), a beta (PRKAB1 or PRKAB2) and a gamma non-catalytic subunits (PRKAG1, PRKAG2 or PRKAG3) (PubMed:21680840). Interacts with FNIP1 and FNIP2 (PubMed:17028174, 18403135, 18663353).
