Recombinant anti-Aβ42 intrabody in fusion with +36GFP works within cells to bind intracellur Aβ42, expressed in E. coli cells. The intrabodies, one with an endoplasmic reticulum (ER) targeting signal and one devoid of a targeting sequence, were assessed in cells harboring a doxycycline (Dox)-regulated mutant human amyloid precursor protein Swedish mutant (hAPPswe) transcription unit for their abilities to prevent Aβ peptide egress.

GFP with a theoretical net charge of +36 (+36 GFP) penetrated several different mammalian cell lines at very low (nanomolar) concentrations. Cationic peptides and proteins have been observed to penetrate cells, and both natural and synthetic cell-penetrating peptides are widely used as delivery vehicles. However, whereas a covalent link between the cell-penetrating peptide and its cargo is typically required.
