What is the toxin component?

The toxin component is Gelonin, a ribosomal-inactivating protein purified from plant seeds. It inhibits protein synthesis by damaging the 60S subunit of mammalian ribosomes after entering the cell.

How does it enter the cell?

The immunotoxin binds selectively to the CD5 antigen on T cells. Once bound, it enters the cells through receptor-mediated endocytosis, allowing the toxin to exert its effect.

anti-CD5 immunotoxin H65 (scFv)-Gel

This immunotoxin was achieved by conjugating/fusing the Anti-CD5 scFv to gelonin. It was expressed in CHO and purified with affinity chromatography. The immunotoxin selectively binds to T cells which express the CD5 antigen. Once bind, the toxin enters cells by receptor-mediated endocytosis, then gelonin inhibits protein synthesis by damaging the 60S subunit of mammalian ribosomes. It was designed for treatment of T cell leukemias or lymphomas, graft-versus-host diseases, and autoimmune diseases.

Gelonin is a glycoprotein (M.W. approximately 29-30, 000 Kd) purified from the seeds of Gelonium multiforum and belongs to a class of potent ribosomal-inactivating plant toxins. Gelonin, like abrin and ricin, inhibits protein synthesis by damaging the 60S subunit of mammalian ribosomes.
