SmartAb™ Recombinant Anti-CD5 pH-dependent Antibody (Clone Zolimomab)

The anti-CD5 pH-dependent antibody (clone Zolimomab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD5. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD5 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD5 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD5 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD5 is a transmembrane protein expressed on T-cells and a subset of B-cells, where it serves as a negative regulator of immune cell activation. It helps to modulate T-cell receptor signaling and is also a marker for a type of chronic lymphocytic leukemia.
