Mouse Anti-EPHA2 Recombinant Antibody (clone EA5) (CAT#: HPAB-0339-YC)

Figure 1 EphA2 antibodies promote tyrosine phosphorylation and degradation of EphA2 in MDA-MB-231 cells.
Monolayers of MDA-MB-231 cells were incubated in the presence of presence of 30 μgml EA5 or EA2 or a control for 24 hours at 37°C. Cell lysates were then resolved by SDS PAGE and subjected to western blot analysis with an EphA2- specific antibody. EphA2 protein levels decrease with antibody incubation. The relative mobility of molecular mass standards is shown on the left of each blot.
Figure 2 EphA2 antibodies inhibit malignant tumor cell growth in vitro.
Purified EphA2 antibodies were incubated with both malignant and benign tumor cells for 7 days at 37°C in soft agar. A549 malignant lung cancer cells were incubated with either 10 μgml or 2.5 μgml of EA5 or EA2 monoclonal antibodies or a control (PBS). All amounts of antibodies used inhibited cell growth in soft agar.

MDA-MB-231 breast cancer cells were implanted into athymic mice. After the tumors had grown to an average volume of 100mm , mice were administered 6 mgkg of the indicated antibody or negative control (PBS or 1A7 antibody) intraperitoneally twice a week for 3 weeks. Tumor growth was assessed and expressed as a ratio of the tumor volume divided by initial tumor volume (100 mm3).
