ELISA OD values are on the vertical axis, elution fractions on the horizontal axis and the individual and joint chain refoldings
are listed on the angular axis. The peak reactivity corresponds to the peak of εγ chain elution. Detection of FN18 binding to the bottom layer is detected by anti-mouse IgG1 mAb conjugated with alkaline phosphatase.

  • Enzyme-linked Immunosorbent Assay
    The CD3 ectodomain or their homo- and heterodimers at 10 μg/ml in PBS were adsorbed to Costar 96-well EIA/Radioimmunoassay flat bottom plates (Corning Inc., NY, Cat# 3590) overnight at 4 centigrade or 2 h at room temperature and then washed three times with PBS containing 0.1% Tween 20. Wells were then incubated for 1 h at room temperature with 3% gelatin in PBS to prevent non-specific binding to the plastic. The wells were then washed as earlier, and incubated for 1 h with FN18 or SP34 mAb (10μg/ml) in PBS containing 0.1% Tween 20. Wells were washed and incubated for another hour with 100 μl goat anti-mouse IgG1 mAb conjugated with alkaline phosphatase(1:1000) or alkaline phosphatase conjugated Protein A for SP34. Wells were washed and alkaline phosphatase substrate (Pierce) was added following the manufacturer's directions. After 30 min, color development was stopped with NaOH and the optical density (OD) was measured with an Enzyme-linked Immunosorbent Assay reader using a 405 nm filter. Each sample was performed in triplicate. Soluble competition Enzyme-linked Immunosorbent Assay was performed by binding εγ heterodimer to the plastic as above and blocking and washing as above. FN18 antibody at 2 μg/ml was mixed with various concentrations competitor CD3 chains. Detection of FN18 binding to the solid phase was done as above. Solid phase competition Enzyme-linked Immunosorbent Assay was performed by mixing various ratios of CD3 chain competitors with εγ heterodimer and attaching these mixtures to the plates. FN18 binding to the solid phase was detected as before. UCHT1 served as an IgG1 isotype control for FN18.
