The anti-CD6 pH-dependent antibody (VS-0825-YC65) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD6. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD6 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD6 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD6 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD6 is a co-stimulatory receptor expressed on T-cells and a subset of B-cells that plays a role in cell-cell adhesion and T-cell activation. It binds to ALCAM (CD166) and contributes to the formation of the immunological synapse.
