The anti-CD74 pH-dependent antibody (VS-0825-YC68) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to both human and monkey CD74. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD74 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD74 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD74 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. CD74, also known as the invariant chain, is a transmembrane protein involved in the antigen processing and presentation pathway. It is expressed in both human and monkey B-cells and antigen-presenting cells, playing a critical role in the proper folding and transport of MHC class II molecules.
