The anti-CD79B pH-dependent antibody (VS-0825-YC69) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CD79B. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CD79B under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CD79B at pH 6.0. This pH-sensitive antibody retains its binding affinity for CD79B on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CD79B is a component of the B-cell receptor (BCR) complex, serving as a signaling subunit alongside CD79A. Upon antigen binding to the BCR, CD79A and CD79B transduce the signal into the cell, leading to B-cell activation.
