The anti-CFD pH-dependent antibody (VS-0825-YC79) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CFD. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CFD under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CFD at pH 6.0. This pH-sensitive antibody retains its binding affinity for CFD on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CFD, also known as Factor D, is a serine protease that is a key component of the alternative pathway of the complement system. It cleaves factor B to initiate the cascade, playing a critical role in innate immunity.
