CFD: The Rate-Limiting Switch for the Alternative Pathway

Factor D (CFD) is a distinctive circulating serine protease that is found only as an already-active enzyme, at a very low circulating concentration. The only known function of this enzyme is to cleave Complement Factor B (CFB) after CFB has been recruited and bound to C3b. This proteolytic action is the key, rate-limiting step that generates the active C3 convertase (C3bBb). Since CFD is the "on-switch" for the entire alternative pathway amplification loop, CFD is an attractive therapeutic target. CFD is also secreted from adipocytes (fat cells) and plays a role in the low-grade chronic inflammation seen in obesity.

Background

Factor D is a protein which in humans is encoded by the CFD gene. Factor D is involved in the alternative complement pathway of the complement system where it cleaves factor B.
