The anti-CSF1 pH-dependent antibody (VS-0825-YC83) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CSF1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CSF1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CSF1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for CSF1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CSF1, or Macrophage Colony-Stimulating Factor, is a cytokine that promotes the proliferation, differentiation, and survival of monocytes and macrophages. It plays a key role in inflammation and is a crucial regulator of the monocyte-macrophage lineage.
