The anti-GPC3 pH-dependent antibody (clone Codrituzumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human GPC3. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for GPC3 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from GPC3 at pH 6.0. This pH-sensitive antibody retains its binding affinity for GPC3 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human GPC3, or Glypican-3, is a heparan sulfate proteoglycan that is anchored to the cell membrane. It is a marker for hepatocellular carcinoma and is being investigated as a therapeutic target for this liver cancer.
