The anti-ICAM1 pH-dependent antibody (VS-0825-YC159) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human ICAM1. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for ICAM1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from ICAM1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for ICAM1 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human ICAM1, or Intercellular Adhesion Molecule 1, also known as CD54, is a cell surface glycoprotein that is involved in cell-cell adhesion. It is expressed on endothelial cells and immune cells and is critical for the trafficking of leukocytes to inflammatory sites.
