The anti-IFNAR1 pH-dependent antibody (clone MEDI546) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human interferon (alpha, beta, and omega) receptor 1 (IFNAR1). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IFNAR1 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IFNAR1 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IFNAR1 on tumor cells while exhibiting negligible binding to the same receptor on normal cells at physiological pH. IFNAR1 is a type I membrane protein that, in conjunction with IFNAR2, forms the receptor complex for Type I interferons. This receptor plays a critical role in immune function by initiating the JAK-STAT signaling pathway, which mediates a robust antiviral and immune response.
