Provided is a immunoglobulin that binds IgE with high affinity. The antibody is capable of inhibiting cells that express membrane-anchored IgE. Also, the antibody is useful for treating IgE-mediated disorders, including allergies and asthma.

Histamine release was performed as previously published, Inhibition of ragweed-induced histamine release was measured by coincubating anti-IgE mAbs with lOOng/ml ragweedspecific
IgE for 2 h at 37°C followed by a 30-min challenge with 100 ng/ml ragweed antigen. Data represents I of 3 experiments.

Mutants were prepared [4], transiently expressed and quantified by ELISA. Binding of CHO cells to 1 ug/ml IgE mutants was measured by immunofluorescence and analyzed by flow cytometry, and binding to anti-IgE mAbs was measured by ELISA. Values are the average of duplicate determinations from 1 of 3 experiments.

Human lung strips were incubated with a 1 : 10 dilution of human ragweed serum for 20 hours in the presence of an equimolar concentration of MaE11 or a IO-fold excess concentration of control anti-IgE mAb, MaEl. The lo-fold excess concentration of MaEl inhibited histamine release (A) and tissue contraction (B) in response to AgE challenge by about 50%. In contrast, equimolar concentrations of MaEll completely blocked both histamine release (A) and tissue contraction (B) in response to AgE challenge. Results are expressed as the mean ± SEM of five separate experiments.
