The anti-MIF pH-dependent antibody (VS-0825-YC252) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human MIF. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MIF under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MIF at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MIF (Macrophage Migration Inhibitory Factor) is a pleiotropic cytokine that functions as a pro-inflammatory mediator. It plays a role in regulating immune responses and inflammation and has been implicated in the development and progression of cancer.
