The anti-RTN4 pH-dependent antibody (VS-0825-YC298) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human RTN-4. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for RTN4 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from RTN4 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human RTN-4 (Reticulon 4), also known as Nogo, is a membrane protein that acts as a potent inhibitor of axon regeneration in the central nervous system. It is a key player in spinal cord injury and is implicated in demyelinating diseases.
