The anti-TNFRSF10A pH-dependent antibody (clone Mapatumumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human TNFRSF10A. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for TNFRSF10A under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from TNFRSF10A at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human TNFRSF10A, also known as TRAIL-R1 or DR4, is a death receptor that binds to the TRAIL ligand to induce apoptosis in target cells. It is a promising therapeutic target for cancer due to its role in triggering programmed cell death.
