The anti-VEGF pH-dependent antibody (clone 4D3) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human VEGF. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for VEGF under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from VEGF at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human VEGF (Vascular Endothelial Growth Factor) is a signaling protein and the master regulator of angiogenesis. It is a key mediator of tumor growth and is a major therapeutic target for a variety of cancers.
