ACLY: The Central Link Between Carbohydrate and Lipid Metabolism

ATP Citrate Lyase (ACLY) is an enzyme found in the cytosol of cells. This enzyme is responsible for converting citrate into acetyl-CoA, which is the main precursor for fatty acid and cholesterol synthesis. This reaction transports carbon atoms from glucose (obtained through glycolysis and the Krebs cycle) into lipids. ACLY activity is upregulated in cancer cells, which require an increased lipid supply for membrane synthesis to drive proliferation. ACLY-derived acetyl-CoA is also the main source for cholesterol synthesis in the liver. Inhibition of ACLY is therefore an established means to reduce cholesterol levels and an emerging strategy to deprive tumor cells of building blocks.

Background

ATP citrate lyase is the primary enzyme responsible for the synthesis of cytosolic acetyl-CoA in many tissues. The enzyme is a tetramer (relative molecular weight approximately 440, 000) of apparently identical subunits. It catalyzes the formation of acetyl-CoA and oxaloacetate from citrate and CoA with a concomitant hydrolysis of ATP to ADP and phosphate. The product, acetyl-CoA, serves several important biosynthetic pathways, including lipogenesis and cholesterogenesis. In nervous tissue, ATP citrate-lyase may be involved in the biosynthesis of acetylcholine. Multiple transcript variants encoding distinct isoforms have been identified for this gene.
