CD40LG is a protein that is primarily expressed on activated T cells and is a member of the TNF superfamily molecule. It binds to CD40 on antigen-presenting cells (APCs), depending on the target cell type resulting in many effects. In total CD40LG has three binding partners: CD40, α5β1 integrin and αIIbβ3. CD40LG acts as a costimulatory molecule and is particularly important for a subset of T cells called T follicular helper cells. In T follicular helper cells, CD40LG promotes B cell maturation and function by conjugating B cell surface CD40 and promoting cell-cell communication. Defects in this gene result in the inability to undergo immunoglobulin class switching and are associated with the high IgM syndrome. The lack of CD40LG also prevents the formation of germinal centers, thus inhibiting antibody affinity maturation, which is an important process in the adaptive immune system. The CD40 ligand is mainly expressed on activated CD4+ T lymphocytes but also in a soluble form. Although CD40LG was originally described on T lymphocytes, its expression has been found on a variety of cells, including platelets, mast cells, macrophages, basophils, NK cells, B lymphocytes, and non-hematopoietic cells (smooth muscle cells). , endothelial cells and epithelial cells).
