Anti-B2M Antibody Prodrug, Protease Activated (B2M-1D8)

The Anti-B2M Antibody Prodrug, Protease Activated (B2M-1D8) is an antibody prodrug comprised of a recombinant human antibody targeting human B2M, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to B2M protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to B2M effectively. B2M (Beta-2 Microglobulin) is a component of the MHC Class I complex, which is essential for presenting tumor antigens to cytotoxic T-cells. Loss-of-function mutations in B2M are common in advanced cancers (especially melanoma and colorectal cancer) treated with immunotherapy. This loss results in the inability to display antigens, effectively making the tumor cell invisible to T-cell recognition and attack.
