The anti-CSF1R pH-dependent antibody (VS-0825-YC84) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human CSF1R. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for CSF1R under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from CSF1R at pH 6.0. This pH-sensitive antibody retains its binding affinity for CSF1R on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human CSF1R is a tyrosine kinase receptor that binds to CSF1, transducing signals that are essential for the survival and differentiation of monocytes and macrophages. It is a major therapeutic target for a variety of diseases, including cancer and inflammatory disorders.
