The anti-EPHA3 pH-dependent antibody (VS-0825-YC115) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human EPHA3. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for EPHA3 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from EPHA3 at pH 6.0. This pH-sensitive antibody retains its binding affinity for EPHA3 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human EPHA3 is a receptor tyrosine kinase of the Ephrin family that plays a role in cell adhesion and migration. It is involved in various developmental processes, including axon guidance and angiogenesis, and its dysregulation is linked to cancer.
