The anti-FGF2 pH-dependent antibody (VS-0825-YC129) targets human FGF-2 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-FGF2 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced FGF2 clearance. The antibody operates in a cyclical manner, binding FGF2 in the extracellular fluid and releasing it in acidified endosomes. Human FGF-2, or Fibroblast Growth Factor 2, is a pleiotropic growth factor involved in a wide range of biological processes, including cell proliferation, angiogenesis, and wound healing. It is a key mediator of vascular development and is implicated in various cancers.
