SmartAb™ Recombinant Anti-IL6R pH-dependent Antibody (VS-0825-YC207)

The anti-IL6R pH-dependent antibody (VS-0825-YC207) targets human IL6R in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-IL6R acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced IL6R clearance. The antibody operates in a cyclical manner, binding IL6R in the extracellular fluid and releasing it in acidified endosomes. IL-6R (Interleukin-6 Receptor) is a cell-surface receptor that binds to the cytokine IL-6, playing a key role in inflammation, the acute phase response, and B cell maturation. Targeting this receptor is a therapeutic strategy for inflammatory diseases like rheumatoid arthritis.
