The anti-IL6R pH-dependent antibody (VS-0825-YC16) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human Interleukin-6 Receptor (IL-6R). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IL-6R under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IL-6R at pH 6.0. This pH-sensitive antibody retains its binding affinity for IL-6R on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IL-6R is a cell-surface receptor that binds to the cytokine IL-6, playing a key role in inflammation, the acute phase response, and B cell maturation. Targeting this receptor is a therapeutic strategy for inflammatory diseases such as rheumatoid arthritis.
