The anti-MMP9 pH-dependent antibody (VS-0825-YC256) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human, cynomolgus, and rat MMP9 protein. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MMP9 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MMP9 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MMP9, also known as Gelatinase B, is a protease that degrades components of the extracellular matrix. It is a key mediator of tissue remodeling, inflammation, and is a crucial player in tumor angiogenesis and metastasis.
