The anti-NTRK2 pH-dependent antibody (clone C20) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human, cynomolgus, and mouse Tropomyosin receptor kinase B (TrkB). By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for NTRK2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from NTRK2 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. NTRK2, also known as TrkB, is a receptor tyrosine kinase that binds to Brain-Derived Neurotrophic Factor (BDNF). It plays a crucial role in neuronal survival and synaptic plasticity, and its dysregulation is linked to mood disorders and certain cancers.
