SmartAb™ Recombinant Anti-PCSK9 pH-dependent Antibody (Clone Lodelcizumab)

The antibody binds to human PCSK9 with pH-dependent antigen binding properties. The antibody preferred PCSK9 binding under the acidic pH relative to physiological pH by introducing His mutations, which could act as a pH-switch. By employing yeast surface display with a pH-dependent screening pressure, mAbs having markedly increased dissociation from PCSK9 at pH 6.0 were identified. The pH-sensitive antibody maintained a binding affinity on tumor cells, while having negligible binding to PCSK9 expressed on normal cells under physiological pH. Human PCSK9, or Proprotein convertase subtilisin/kexin type 9, is an enzyme that regulates the level of LDL cholesterol in the blood. It binds to and promotes the degradation of the LDL receptor, resulting in higher blood levels of LDL cholesterol.
