SmartAb™ Recombinant Anti-SIRPA pH-dependent Antibody (Clone AM4-1)

The anti-SIRPA pH-dependent antibody (clone AM4-1) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human SIRPA. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SIRPA under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SIRPA at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human SIRPA (Signal-Regulatory Protein Alpha), also known as CD172a, is a transmembrane protein expressed on myeloid cells, such as macrophages. It acts as a receptor for CD47, and this interaction delivers a "don't eat me" signal that prevents phagocytosis. Disruption of this pathway is a major strategy in cancer immunotherapy.
