The anti-STAT3 pH-dependent antibody (VS-0825-YC316) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human STAT3. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for STAT3 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from STAT3 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human STAT3 (Signal Transducer and Activator of Transcription 3) is a transcription factor involved in cell growth, proliferation, and survival. Constitutive activation of STAT3 is a key oncogenic feature in many cancers, making it a major therapeutic target.
