The anti-SYK pH-dependent antibody (VS-0825-YC318) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human SYK. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for SYK under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from SYK at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human SYK (Spleen Tyrosine Kinase) is a non-receptor tyrosine kinase that is crucial for B-cell and Fc receptor signaling. It is a key player in immune responses and is a therapeutic target for autoimmune diseases and B-cell lymphomas.
