BRD4: A Master Regulator of Oncogenic Transcription

A BET family member, Bromodomain-containing protein 4 (BRD4) is one of the most prolific cancer targets in the industry. This master transcriptional co-activator plays a key role in the transcriptional machinery. BRD4 is a chromatin “reader” that recognizes acetylated histones at enhancers and promoters and recruits the positive transcription elongation factor b (P-TEFb) and other components of the transcriptional machinery, to function as a scaffold. BRD4 plays a key role in regulating the expression of critical oncogenes, including MYC. It's essential to the abnormal transcriptional circuits at the heart of many leukemias, lymphomas, and solid tumors. BRD4 is a master regulator of oncogenic transcription, and blocking its function with small molecule BET inhibitors has been a hot oncology target in recent years.

Background

The protein product of this gene is homologous to the murine protein MCAP, which binds to chromosomes during mitosis, and to the human RING3 protein, which is a serine/threonine kinase. All three of these proteins have two bromodomains, a conserved sequence motif that may be involved in targeting these proteins to chromatin. This gene was also identified as the chromosome 19 target of translocation t(15;19)(q13;p13.1), which is responsible for an upper respiratory tract carcinoma in young individuals. Two alternatively spliced transcript variants have been described.
