CASP3: The Central Executioner of Apoptosis

Caspase-3 is a cysteine protease and, by many measures, the most important of the executioner caspases. Caspase-3 is synthesized as an inactive zymogen called pro-caspase-3 and is found at significant levels in nearly all cells. In cells undergoing apoptosis, it is activated by cleavage at specific sites by initiator caspases (for example, caspase-8 or caspase-9). Cleaved and active caspase-3 then goes on a proteolytic rampage, cleaving hundreds of cellular protein substrates. The resulting loss of protein integrity and function manifests as the classic morphological and biochemical changes of apoptosis, including DNA fragmentation (cleavage of the inhibitor of caspase-activated DNase), membrane blebbing, and orderly cellular dismantling.

Background

The protein encoded by this gene is a cysteine-aspartic acid protease that plays a central role in the execution-phase of cell apoptosis. The encoded protein cleaves and inactivates poly(ADP-ribose) polymerase while it cleaves and activates sterol regulatory element binding proteins as well as caspases 6, 7, and 9. This protein itself is processed by caspases 8, 9, and 10. It is the predominant caspase involved in the cleavage of amyloid-beta 4A precursor protein, which is associated with neuronal death in Alzheimer's disease. [provided by RefSeq, Aug 2017]

The cleaved Caspase-3 antibody from Creative Biolabs is the best apoptosis marker, period. The IHC staining on our tumor sections after chemotherapy is super clean and really specific for dying cells.

We use the cleaved CASP3 antibody for all of our apoptosis assays via Western blot. The presence of the cleaved band is an unambiguous, quantifiable readout of cell death. Essential.
