Background

C-X-C chemokine receptor 4 (CXCR4) is a G protein-coupled receptor (GPCR) binding the chemokine CXCL12 (SDF-1α). Expressed on hematopoietic stem cells, immune cells, and tumor cells, it regulates cell migration, homing, survival, and proliferation. Overexpressed in multiple cancers (AML, NHL, breast, prostate), CXCR4 promotes tumor metastasis by guiding cells to CXCL12-rich tissues (bone marrow, lungs). It also contributes to chemoresistance and cancer stem cell maintenance. Targeted therapies (plerixafor, monoclonal antibodies) block CXCR4-CXCL12 signaling, inhibiting metastasis and enhancing chemotherapy efficacy, with applications in hematologic and solid tumors.
