Human Anti-CXCR4 Recombinant Antibody (clone Ab124) (CAT#: HPAB-1842-FY)

This product is a recombinant antibody that recognizes CXCR4. The monoclonal antibody Ab124 reacts with the epitope of CXCR4. Ab124 can inhibit SDF-1 induced chemotaxis in human breast cancer line MDA-MB-231.

ELISA-binding assays were performed to measure anti-CXCR4 scFv (Ab124; Ab125) binding to PC-3 breast cancer cells.

Flow cytometric analysis was performed using a three-step procedure. Cells were first incubated with Ab16, Ab124, or Ab125 (10 μg/ml) or without scFv, followed by anti-HSV Tag mAb (0.5 μg/ml), then fluorescent antimouse IgG antibody.

The effects of Ab124 and Ab125 on calcium mobilization in PCa cells was determined. PC-3 and LNCaP cells were preincubated with Ab16, Ab124, or Ab125 (10 μg/ml) for 30 min, then exposed to CXCL12 (100 ng/ml) for measurement of calcium flux.

Chemotaxis assays were performed to determine the inhibitory effects of Ab124 and Ab125 on CXCL12-induced migration of PCa cells and control T cells. PC-3 cells were preincubated with Ab16, Ab124, or Ab125 (10 μg/ml) for 30 min at 37°C, then incubated in Transwell migration chambers containing CXCL12 (400 ng/ml) for 18–24 h.
