Anti-Human DKK1 Recombinant Antibody (RH2-18)

This antibody, specific for Dkk-1, can be potentially used as an inhibitor of the osteoanabolic Wnt/LRP5 signaling pathway for treating bone disorders which results in a loss in bone, for example, osteoporosis.

Figure 1 shows Eu-Dkk-1 binding to HEK293hLrp5 cells and inhibitory activity of anti-Dkk-1 antibodies RH2-10, RH2-18, RH2-31, RH2-59, and RH2-80 at various concentrations.

ELISA

Figure 2 shows the result for RH2-18 antibodies retitrated using an extended dose range. The results shows that for this assay format the RH2-18 antibodies had an effective dose of about 5 nM.

FuncS

Figure 3 shows that distal femur bone mineral density (BMD) was increased 5.2 to 8.7% in a dose effect fashion by RH2-18 antibodies in the dose range 0.5 to 5 mg/kg. Error bars = SEM. N=I l /group.

Figure 4 shows a xenograph model for tumor growth and the effect of the antibody on tumor growth.
