The anti-DKK1 pH-dependent antibody (VS-0825-YC99) targets human DKK1 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-DKK1 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced DKK1 clearance. The antibody operates in a cyclical manner, binding DKK1 in the extracellular fluid and releasing it in acidified endosomes. Human DKK1, or Dickkopf-1, is a secreted protein that acts as an antagonist of the Wnt signaling pathway. It plays a key role in embryonic development and is frequently overexpressed in various cancers, promoting tumor growth and metastasis.
