SmartAb™ Recombinant Anti-BSG pH-dependent Antibody (Clone 5F6)

The anti-BSG pH-dependent antibody (clone 5F6) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human BSG. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for BSG under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from BSG at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human BSG (Basigin), also known as CD147, is a transmembrane glycoprotein involved in cell adhesion and is a key regulator of matrix metalloproteinases (MMPs). It is often overexpressed in cancer, where it promotes tumor invasion and metastasis.
