The Anti-HLA-A Antibody Prodrug, Protease Activated (HYB3) is an antibody prodrug comprised of a recombinant human antibody targeting human HLA-A, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to HLA-A protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to HLA-A effectively. HLA-A (Human Leukocyte Antigen A) is a core component of the MHC Class I complex found on nearly all human cells and is essential for presenting tumor antigens to cytotoxic T-cells. Loss or significant downregulation of HLA-A expression by tumor cells is a common immune evasion mechanism, rendering the cancer effectively invisible to the immune system.

- The HLA class I antigen is formed by a heterodimer of a 45-KD α comprising three domains (α1, α2, α3), and a 12-KD β2 microglobulin. The main role of the HLA molecule is to present CD8+T cells with antigenic peptides formed from about eight to ten amino acids and produced inside cells.
