The anti-IGF2 pH-dependent antibody (clone Dusigitumab) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human IGF2. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for IGF2 under acidic conditions compared to physiological pH. Using a yeast surface display system with pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from IGF2 at pH 6.0. This pH-sensitive antibody retains its binding affinity for IGF2 on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human IGF2, or Insulin-like Growth Factor 2, is a hormone that is structurally similar to insulin and IGF1. It plays a key role in embryonic development and is also involved in the growth of certain cancers.
